Greetings, All!

I hope you all had a lovely weekend and are looking forward to all that this final week in March has to offer! Seems we have lots going on as we embark on this last stretch of the semester, so be sure to stay focused, but to also give yourself permission to take breaks and do fun things in between all of the hustle and bustle of this busy time of year. Swing by the office any time for candy, a hot drink, and a fun fact of the day as needed. 😊 I just returned from a wonderful trip to Japan and hope to bring in a few snacks/treats from my travels sometime later this week.

Monday, March 27th:

  *   Deadline for registering for the SWE Dinner later this week! (See attached flyer.)
  *   Deadline for speaking with the Ivanhoe International Center about applying to study abroad in Norway at OsloMet.
  *   6:30PM – The Future Health Science Professionals (FHSP) is having a meeting in CBEC 3304 to talk about the needs of pre-health students on campus. Points raised at this discussion will be carried forward to the newly resurrected Pre-Health Advisory Committee, formed to help support pre-health students across campus. If you are interested in a career as a healthcare professional, I highly encourage you to attend this meeting and voice your concerns/needs.

     *   Abstract: Macropinocytosis is the process of cell drinking in which plasma membrane rufflesextend, curve, and return to the plasma membrane surface to form large intracellularorganelles called a macropinosome. This complex process can be carefully understoodthrough the constant advancements in multidimensional fluorescence microscopy,specifically lattice light sheet (LLS), however there follows a demand for datavisualization tools capable of displaying the complex cellular processes. While LLSmicroscopes produce stunning movies of cellular activity with high spatial and temporalresolution, they have been slow to be widely adopted. Experiments on this instrumentrequire understanding fluorescence microscopy, cell biology, data management,computer science, and most importantly presenting 5 dimensions of data (x, y, z, t, λ) intoa scientifically accurate 2-dimensional manuscript. The first objective focused on how todevelop an experiment for LLS microscopy as many traditional experiments requireunique preparations and considerations. Experimental development was followed by adiscussion of the limitations, and applications of the different visualization tools thatwere developed including volumetric intensities, orthogonal planes, isosurfaces, andmesh overlays. Furthermore, a non-programing based step-by-step protocol was createdfor using ChimeraX for LLS data visualization. These techniques were developed tounderstand macropinocytosis, a process where actin-rich plasma membrane ruffles forminto large intracellular organelles, in a phosphatidylinositol 3-kinase (PI3K)-coordinatedmanner. The visualization tools were used to show that macropinosomes were formedthrough multiple ruffling morphologies and that the majority formed through collisions ofadjacent PI3K-rich ruffles. Furthermore, we showed that PI3K activity begins earlythroughout the ruffle volume and continued to increase until peak activity concentrated atthe base of the ruffle after macropinosome closure. Additionally, through pharmacologicinhibition of PI3K activity we showed there was little effect on the rate and morphologyof membrane ruffling. Finally, we demonstrated that early production of 3′-phosphoinositides within ruffles plays a minor role in regulating their morphologyhowever, 3′-phosphoinositides are critical for sealing ruffles into macropinosomes.Through the development and application of LLS visualization techniques these dataindicate that local PI3K activity is amplified in ruffles and serves as a primingmechanism for closure and sealing of ruffles into macropinosomes.
